Acyclopyrimidine<i>C</i>-nucleosides. Synthesis of acyclopseudoisocytidine and its derivatives

with dimethylformamide dimethyl acetal gave 1,3-dimethylacyclopseudouridine (VI) which was subsequently converted to acyclopseudoisocytidine (VII). 1-Methyl derivatives V and VIII were obtained from the selective methylation of IV and VII, respectively. 5-Hydroxymethyluracil also reacted with 2-mercaptoethanol to give the sulfide derivative (IX).
由5-羟甲基尿嘧啶(I)合成无环伪古尿苷(IV),无环伪异胞苷(VII)及其1-甲基衍生物(V和VIII)。由5-羟甲基尿嘧啶(I)与乙二醇在酸性条件下的缩合反应可方便地制备无环假双胍(IV)。化合物Ⅳ也可以由5-氯甲基尿嘧啶制备,但是,发现该方法不如直接缩合法。IV用二甲基甲酰胺二甲基乙缩醛的甲基化得到1,3-二甲基acyclopseudouridine(VI),其随后转化为无环假异胞苷(VII)。1-甲基衍生物V和VIII分别由IV和VII的选择性甲基化获得。5-羟甲基尿嘧啶也与2-巯基乙醇反应,得到硫化物衍生物(IX)。
