    摘要:
    生物有效的有效ACAT抑制剂可通过(i)减少饮食中胆固醇的吸收,(ii)减少极低密度脂蛋白从肝脏向血浆的分泌以及(iii)防止转化而对动脉粥样硬化产生有益作用巨噬细胞进入泡沫细胞。我们已经发现,含有4,5-二苯基-1H-咪唑-2-基部分的甲羟戊酸酯衍生物2在体外抑制大鼠肝微粒体ACAT,并在胆固醇喂养的大鼠中产生明显的降胆固醇作用。2的类似物的结构活性关系表明,4,5-二苯基-1H-咪唑部分是抑制大鼠微粒体ACAT的药效基团。

    The present disclosure relates to the technical field of biochemical engineering and particularly discloses a preparation method for (R)-3-hydroxyl-5-hexenoate. In the method of the present disclosure, the (R)-3-hydroxyl-5-hexenoate is prepared by catalytic reduction of 3-carbonyl-5-hexenoate by ketoreductase with 3-carbonyl-5-hexenoate as the substrate. The amino acid sequence of ketoreductase is shown in SEQ ID NO.1. In the present disclosure, the (R)-3-hydroxyl-5-hexenoate having a very high chiral purity is obtained by asymmetric reduction by ketoreductase as the biocatalyst. The present disclosure has the advantages of easy operation, mild reaction conditions, high reaction yield and good practical industrial application value.

    A method is provided which biosynthesizes a PHA having an epoxy group in a side chain terminal with improved physicochemical properties. Specifically, a method of producing a polyester containing an epoxy group in a side chain thereof using 1-alkene as a raw material is provided which comprises the steps of bringing 1-alkene into contact with a microorganism having an ability to uptake 1-alkene and convert it to a polyester and allowing the microorganism to convert the 1-alkene into a polyester containing an epoxy group in a side chain thereof. Further, a method of producing a crosslinked polymer is provided which comprises reacting the polyester obtained by the above mentioned method with a diamine compound.

    Provided is a method of preparing polylactate (PLA) or a copolymer thereof using a mutant microorganism in which a gene participating in a coenzyme A (CoA) donor- and lactate-producing pathway is genetically manipulated to increase the productivity of a CoA donor and lactate. Amounts of the CoA donor and the lactate are simultaneously increased in a microbial metabolic pathway to enable effective biosynthesis of PLA and a hydroxyalkanoate-lactate copolymer having a high content of lactate, which is industrially useful.
