    New Metallodendrimers Containing an Octakis(diphenylphosphino)-Functionalized Silsesquioxane Core and Ruthenium(II)-Based Chromophores
    摘要:
    A new class of surface-modified dendrimers has been prepared by reactions of 8 equiv of the terpyridine-functionalized polyether monodendrons with a polyhedral oligomeric silsesquioxane (POSS) core. Subsequent reactions of these spherically shaped organic dendrimers with Ru(II)-based precursors afford photo- and redox-active metallodendrimers. These new dendrimers have been characterized using a combination of mass spectral analysis (MALDI-TOF/MS, ESI/MS, and FAB/MS), nuclear magnetic resonance (H-1, C-13, Si-29, and (31)p{H-1} NMR), photophysical analyses (electronic absorption, emission, excited-state lifetime, and quantum yield) and electrochemical measurement (cyclic voltammetry). Specifically, (31)p{H-1} NMR is used to monitor the completion of reactions and the purity of dendrimers and metallodendrimers. These new metallodendrimers exhibit large extinction coefficients that coincide with the number of peripheral Ru(II)-based chromophores. With the use of (-CH2-Ph-tpy)Ru-II(bpy)(2) type of chromophores, all metallodendrimers are found emissive at room temperature, with lifetimes in the range of 605-890 ns. Photophysical data also indicate similar steady-state emission maxima and single-exponential decay kinetics for all metallodendrimers, and the observed overall quantum yields of the G1, G2, and G3 metallodendrimers are found to be 14, 20, and 7 times higher than that of the monomeric model complex (CH3-Ph-tpy)Ru(bpy)(2)(PF6)(2). Electrochemical studies reveal the presence of surface-confined species, in addition to the ligand-centered and metal-centered redox processes.

    A terpyridine derivative represented by general formula (I) (wherein R represents hydrogen, (un)substituted phenyl, pyridyl, etc.), which has the effect of promoting the production of nerve growth factors or a neurotrophic factor activity. Hence it can be used as an ameliorant or remedy for the symptoms and diseases accompanying peripheral nerve degeneration which is idiopathic or caused by trauma, chemicals such as alcohol or anticancer drug, inflammation, or metabolism as observed in, for example, diabetes. In addition, it can be used as an ameliorant or remedy for the symptoms and diseases accompanying central nerve degeneration, such as senile dementia of Alzheimer type, cerebrovascular dementia, Down's syndrome, Parkinson's disease or Hantington's chorea, mental and motor function incompetences caused by cerebral ischemia, cerebral infarction, cerebral hemorrhage or head injury, spinal neuroparalysis, and so forth.

    In this contribution, we constructed a biosensor for screening G-quadruplex stabilizers based on the assembly of gold nanoparticles functionalized with guanine-rich (G-rich) ssDNA using a light-scattering technique. We synthesized a series of metal–terpyridine complexes and investigated their affinity for quadruplex-DNA using the biosensor. Using the ratio of intensity of the light-scattering peak (I − I0)/I0, it can intuitively present the sequence of ability of stabilizing G-quadruplex DNA as follows: complex 1 > complex 3 > MTX > complex 2 > complex 5 > complex 6 > complex 7 > complex 4 > methyl green > TO > complex 8 > cisplatin. As our method allows the quantitative analysis of stabilizer affinity, EC50 values obtained are 4.8, 5.3, 6.1, 6.6, 6.7, 18.2, 20.2, 21.5, 32.2, 41.5 and 48.1 μM for complex 1, complex 3, MTX, complex 2, complex 5, complex 6, complex 7, complex 4, methyl green, TO and complex 8, respectively. The results have been verified by G-quadruplex fluorescent intercalator displacement (G4-FID) analysis. The proposed approach is a simple, convenient, intuitive and highly sensitive assay for screening G-quadruplex stabilizers. Our developed biosensor provides a promising tool for screening anticancer drugs.
