(+)-Pyrido[3,4-b]homotropane (PHT) was efficiently constructed in 12 steps from N,N-diisopropylisonicotinamide. The synthesis features (i) RCM reaction in installing the homotropane skeleton, (ii) Snieckus ortho-lithiation of N,N-diisopropylisonicotinamide followed by acylation, (iii) Myer reduction of bulky tertiary amide to alcohol with LiBH3NH2, and (iv) utilization of L-glutamic acid to introduce and establish the requisite stereogenic centers.
(+)-吡rido[3,4-b]齐墩果烷(PHT)通过12步反应由N,N-二异丙基烟酰胺高效合成。该合成过程的特点包括:(i)运用环形缩合反应(RCM)构建齐墩果烷骨架,(ii)对N,N-二异丙基烟酰胺进行Snieckus正交锂化随后进行酰化,(iii)利用LiBH3NH2将庞大的三级酰胺还原为醇,以及(iv)使用L-谷氨酸引入并建立所需的立体生成中心。
