Synthesis of derivatives of 2-amino-2-deoxy-4-O-(α- and β-d-galactopyranosyl)-d-glucose

2-O-(α-cyanobenzylidene)-α- d -galactopyranose (12) gave 1,2-cis- and 1,2-trans-linked disaccharide derivatives in ratios ranging from 3.8:1 to 1:8.5 depending on the conditions of glycosylation. A practical synthesis of the target lactosamine derivative 2 was achieved by the reaction of 5 and 8 in nitromethane in the presence of silver triflate.
摘要用2,3,4,6-四-O-将6-O-乙酰基-3-O-苯甲酰基-2-脱氧-2-邻苯二甲酰亚胺基-β-d-吡喃葡萄糖苷(5)及其三苯甲基醚9的半乳糖基化苯甲酰基-α-d-吡喃半乳糖基溴化物(8),1-O-乙酰基-2,3,4,6-四-O-苯甲酰基-β-d-吡喃半乳糖(13),3,4,6-三-O -苯甲酰基-1,2-O-(α-对甲苯硫基亚苄基)-α-d-吡喃半乳糖(11)和3,4,6-三-O-苯甲酰基-1,2-O-(α-氰基亚苄基) -α-d-吡喃半乳糖(12)根据糖基化条件以3.8∶1至1∶8.5的比例得到1,2-顺式和1,2-反式连接的二糖衍生物。通过在三氟甲磺酸银存在下5和8在硝基甲烷中反应,可以实现目标乳糖胺衍生物2的实际合成。
