A series of substituted 2-aminopyridines was prepared and evaluated as inhibitors of human nitric oxide synthases (NOS). 4,6-Disubstitution enhanced both potency and specificity for the inducible NOS with the most potent compound having an IC50 of 28 nM. A novel and efficient method based on the cross-coupling reactions of pyridine N-oxides with ethers between C(sp2)–H/C(sp3)–H bonds in the presence of TBHP was developed. The strategy provides an alternative approach to the pyridine moiety under transition-metal-free conditions.

A highly efficient protocol for the regioselective dehydrogenative cross-coupling of pyridine-N-oxides with N,N-dialkyl uracils via 2-fold C–H activation has been developed. The resultant uracil substituted pyridine-N-oxide structural motifs is readily deoxygenated, making the present procedure a highly attractive route for the synthesis of 5-pyridine substituted uracils.
