A combined experimental and computational mechanistic study of amide formation from thioacids and azides is described. The data support two distinct mechanistic pathways dependent on the electronic character of the azide component. Relatively electron-rich azides undergo bimolecular coupling with thiocarboxylates via an anion-accelerated [3+2] cycloaddition to give a thiatriazoline. Highly electron-poor
the basis of sequential azide–aryne and azide–alkyne cycloadditions. The key to success was efficient halogen–metalexchange reaction-mediated generation of aryne from ortho-iodoaryl triflates bearing a base-sensitive terminal alkyne moiety, which was achieved using trimethylsilylmethyl Grignardreagent.

chemistry synthesis via CuAAC reaction followed by in situ biological screening were used to discover selective Cdc25 inhibitors. The bioassay results showed that compound M2N12 proved to be the most potent Cdc25 inhibitor, which also act as a highlyselective Cdc25C inhibitor and was about 9-fold potent than that of NSC 663284. Moreover, M2N12 showed remarkable anti-growth activity against the KB-VIN cell

TIPS-acetylene to aryldiazonium tetrafluoroborate salts gives a wide range of azoacetylenes, constituting an underexplored class of stable intermediates. In situ desilylation transiently leads to terminal arylazoacetylenes that undergo copper-catalyzed cycloadditions (CuAAC) with a diverse collection of organoazides. These include complex molecules derivedfrom natural products or drugs, such as colchicine

Bis-triphenylphosphano complexes of copper(I)-carboxylates serve as efficient catalysts for azide-alkyne cycloaddition. The triazole formation takes place straightforwardly at ambient temperature providing a wide variety of products with good yields in the presence of 0.005–0.05% catalyst.
