The present invention relates to aryl sulfones and related compounds that are modulators of ROR-gamma receptors. The invention also provides pharmaceutical compositions comprising these modulators, and methods of modulating ROR-gamma receptors using them. Also provided are methods of using aryl sulfones and related compounds as modulators of ROR-gamma to treat ROR-gamma mediated diseases.

A copper/silver-cocatalyzed protocol for oxidative sulfonylation of hydrazones is demonstrated. A wide range of β-ketosulfones and N-acylsulfonamides are directly synthesized in moderate to good yields. Our work provides a viable method for scalable preparation of β-ketosulfone derivatives that have found wide applications in the pharmaceutical industry.

A novel strategy for the synthesis of sulfonylated lactones via Ag-catalyzed radical addition/cyclization reaction of 1,6-enynes and sodium sulfinates has been developed. The reaction presents high stereoselectivity under mild conditions with C4 prochiral center construction in one step. The ESR experiments and relevant mechanistic studies indicated that a radical pathway should be involved in this
