R(-)-2,3- and S(+)-1,2-O-Isopropylidene-sn-glycerols (4 and 9) are converted into 2-O-arachidonoyl-1-O-stearoyl and 2-O-arachidonoyl-3-O-stearoyl-sn-glycerols (2 and 3, respectively); glycerol is also converted into its racemic 1,2- and its symmetrical 1,3-di-O-linoleoyl derivatives (14 and 17, respectively). The chemical synthesis of naturallyoccurring phosphatidylinositol 3,4,5-trisphosphate [PtdIns(3,4,5)P3] 2, in which the 1- and 2-hydroxy functions of the glycerol moiety are esterified with stearic and arachidonic acid, respectively, is described. The synthesis of three diastereoisomers 41, 42 and 43 of PtdIns(3,4,5)P3 is also reported.

Single enantiomers of PI-4-P, with a range ofsn-2-fatty acid esters, were prepared efficiently. The effects of the degree ofsn-2-unsaturation on the kinetic parameters ofSopB were determined.
