    Derivatives of 1-[2-(4-carboxyphenoxy)ethyl]-3-dodecanoylindole-2-carboxylic acid (4) with modified substituents at the indole-1-position were synthesized and evaluated for their ability to inhibit the arachidonic acid release in human platelets mediated by the cytosolic phospholipase A(2). One of the most active compounds obtained was 26 with an IC50 of 0.44 muM. (C) 2001 Elsevier Science Ltd. All rights reserved.

    New couple: The Cu‐promoted trifluoromethylation of primary and secondary alkylboronic acids with TMSCF3 extends the scope of transition‐metal‐catalyzed trifluoromethylation reactions to sp3‐hybridized carbon centers. It also represents one of the first examples for Cu‐catalyzed CC cross‐coupling reactions of alkylboronic acid derivatives.

    AbstractPerfluoro‐tert‐butylation reaction has long remained a challenging task. We now report the use of 1,1‐dibromo‐2,2‐bis(trifluoromethyl)ethylene (DBBF) as a practical reagent for perfluoro‐tert‐butylation reactions for the first time. Through a consecutive triple‐fluorination process with DBBF and CsF, the (CF3)3C species can be liberated and observed, which is able to serve as a robust nucleophilic perfluoro‐tert‐butylating agent for various electrophiles. The power of this synthetic protocol is evidenced by the efficient synthesis of structurally diverse perfluoro‐tert‐butylated molecules. Multiple applications demonstrate the practicability of this method, as well as the superiority of perfluoro‐tert‐butylated compounds as sensitive probes. The perfluoro‐tert‐butylated product was successfully applied in 1H‐ and 19F‐magnetic resonance imaging (MRI) experiment with an ultra‐low field (ULF) MRI system.
  • Selective thromboxane synthetase inhibitors. 1. 1-[(Aryloxy)alkyl]-1H-imidazoles

    1-(2-Phenoxyethyl)-1H-imidazole was found to be an inhibitor of thromboxane (TxA2) synthetase, but it also inhibited the adrenal cytochrome P-450 enzyme steroid 11 beta-hydroxylase. The preparation of a series of analogues is described, and activity against TxA2 synthetase, PGI2 synthetase, cyclooxygenase, and steroid 11 beta-hydroxylase is discussed. Potency against TxA2 synthetase was increased by introduction of a carboxyl group at a suitable distance from the imidazole ring. A distance of 8.1-8.8 A between N-1 of the imidazole and the carboxyl carbon was found to be optimal. Introduction of a carboxyl group also had the effect of reducing activity against steroid 11 beta-hydroxylase. The most potent and selective compound was found to be 4-[2-(1H-imidazol-1-yl) ethoxy]benzoic acid (14).
