Chemical Modification of PA-48153C, a Novel Immunosuppressant Isolated from Streptomyces prunicolor PA-48153.

(20), 14-methyl (24), 14,14-dibromo-15-nor (25), 8-O-acyl (26-45), 8-O-alkyl (46), 8-O-alkoxycarbonyl (47, 48), and 8-O-carbamoyl (49) derivatives of PA-48153C, a novel immunosuppressant isolated from fermentation products of Streptomyces prunicolor PA-48153, were prepared. These compounds were found to retain the inhibitory activity on the responses of both T and B cells to mitogens. Among them, the
5beta-甲氧基(20),14-甲基(24),14,14-二溴15-nor(25),8-O-酰基(26-45),8-O-烷基(46),8-O制备了分离自短链霉菌PA-48153发酵产物的新型免疫抑制剂PA-48153C的-烷氧羰基(47、48)和8-O-氨基甲酰基(49)衍生物。发现这些化合物保留了对T细胞和B细胞对促细胞分裂剂的响应的抑制活性。其中,C-8己酸酯28对有丝分裂原反应表现出强抑制作用,对EL4细胞的细胞毒性较小,因此被选择用于体内评估。
