A novel five-carbon homologation leading to 3,4-alkadienoic acids by SN2′ reaction of β-ethynyl-β-propiolactone with grignard reagents in the presence of copper(i) catalyst
β-Ethynyl,-β-propiolactone reacts regioselectively with Grignardreagents in the presence of copper(I) catalyst to afford 3,4-alkadienoic acids in high yields. Synthetic utility of this reaction is demonstrated in the simple synthesis of pellitorine.
Liquid crystalline alkane-3,4-dienoates (allenylacetates) have been
synthesized. Most compounds incorporate a heterocyclic 1,3,4-thiadiazole
ring or a pyrimidine ring as a constituent of the rigid core. These axial
chiral allene derivatives were at first obtained as racemic mixtures. Some
of them were also synthesized in enantiomerically enriched form by
enantioselective synthesis. The compounds were investigated by polarizing
microscopy and by differential scanning calorimetry. The three-ring
compounds exhibit broad regions of smectic C-phases. The optically active
three-ring compounds show broad S
c
*-phases with moderate values
of spontaneous polarization.

This invention relates to a process for making a compound of formula I
in the form of a stereoisomer or mixture thereof, wherein R is hydrogen, lower alkyl; X is hydrogen, halo, trifluoromethyl, lower alkyl or lower alkoxy, and the wavy lines represent the a or β configuration with the proviso that when one wavy line is α the other is β, or a pharmaceutically acceptable, non-toxic salt of the compound wherein R is hydrogen; novel intermediates useful for preparing these compounds; processes for making the intermediates; and a stereoisomer of the compound of formula I wherein R is methyl and X is hydrogen and a process for making same.
