Practical synthesis of maleimides and coumarin-linked probes for protein and antibody labelling via reduction of native disulfides

The cellular tracking, detection and sensing of protein or antibody movement are important aspects to advance our understanding of biomolecular interactions and activity. Antibodies modified with fluorescent dyes are also valuable tools, especially in immunology research. We describe here a proof-of-principle study of a new water-soluble coumarin probe with a maleimide thiol-reacting unit to fluorescently tag biomolecules. Highlights include: (1) a convenient water-based preparation of N-substituted maleimides, (2) a one-pot preparation of activated maleimido-esters, and (3) a bio-conjugation protocol for the selenol-promoted reduction of native disulfide bonds and the ‘site-specific’ labelling of antibodies with no significant loss of activity.

The physical properties of blends of olefin polymer and nitrile rubber are improved by the addition of a compatibilizing amount of a block copolymer comprising nitrile rubber- compatibilizing segments and olefin polymer-compatibilizing segments, which segments may themselves be derived respectively from nitrile polymers and olefin polymers containing graft-forming functional groups. In preferred compos- itionsthe nitrile rubber is cured.
Thermoplastic compositions comprising blends of dissimilar plastics and dissimilar cured particulate rubbers in which the plastics are crystalline polyolefin polymer and nylon, and the cured particulate rubbers are hydrocarbon rubber and polar rubber, include a functionalized olefin polymer which is an olefin polymer containing at least one functional group that has a physical or chemical affinity for nylon, to improve the compatibility between the crystalline polyolefin polymer and the nylon.
