play critical roles in biological processes via glycan–protein interactions. However, the structure–functionrelationships of GAGs and glycan-binding proteins are largely unclear due to the lack of structurally well-defined GAGs. Herein, we report a one cycle–one disaccharide unit (OCODU) enzymatic modular assembly strategy for the synthesis of hyaluronan–chondroitin hybrid glycans. With precise employment

Preparation of diverse glycans is essential to glycobiology, while syntheticallyusefulenzymes for this purpose are limited. This study discovered a general tolerance of galactosyltransferases (GalTs) toward uncommon donor UDP-galactosamine. A two-step strategy was devised to harness this property for the facile synthesis of GalNAc-glycosides.

Here, we report a practical sugar nucleotide production strategy that combined a high-concentrated multi-enzyme catalyzed reaction and a robust chromatography-free selective precipitation purification process. Twelve sugar nucleotides were synthesized on a gram scale with a purity up to 98%.
