    Enzymatic synthesis of tritium labelled eicosanoids
    摘要:
    Methods for the synthesis of tritium labelled prostaglandins, thromboxanes, and hydroxy-derivatives of eicosapolyenoic acids using mono-, two-, and three-enzyme systems have been developed. Optimization of the eicosanoid syntheses with respect to the concentrations of enzymes and eicosapolyenoic acid. time, and temperature of incubation are reported. It is demonstrated that a number of compounds can be produced by enzymatic modifications of prostaglandins and thromboxane. Procedures for the preparation of twenty tritium labelled compounds are described.

  • Preparation of high specific activity tritium-labelled leukotriene B4 suitable for radioligand binding assay

    We describe a method of preparation of high specific activity tritium-labelled leukotriene (LT) B4 from [5,6,8,9,11,12,14,15-3H] arachidonic acid (AA; 6.66 TBq/mmol) utilizing a LTB4-synthesizing enzyme system from rat basophilic leukemia (RBL-1) cells. It was shown that both cyclooxygenase inhibitor indomethacin and adenosine 5′-triphosphate induced [3H] AA transformation to [3H] LTB4. In optimized conditions up to 15% of total radioactivity of the incubation mixture was present in [3H] LTB4. A separation of [3H] LTB4 from other labelled C20:4 products was achieved by a three-step reverse phase-high-performance liquid chromatography in methanol- and acetonitrile-based solvent systems. [3H] LTB4 was confirmed to be identical to the naturally occurring LTB4 by a radioligand binding assay using a culture of HF1 cells that express a BLT1 receptor.   • A Methodology for Radiolabeling of the Endocannabinoid 2-Arachidonoylglycerol (2-AG)

    The metabolic intermediate and endocannabinoid signaling lipid 2-arachidonoylglycerol (2-AG) has not been readily labeled, primarily because of its instability toward rearrangement. We now detail a synthetic method that easily gives tritiated 2-AG from [5,6,8,9,11,12,14,15-H-3(N)]arachidonic acid in two steps. We utilized a short chain 1,3-diacylglycerol and proceeded through the "structured lipid" [5 '',6 '',8 '',9 '',11 '',12 '', 14 '',15 ''-H-3(N)]2-arachidonoyl-1,3-dibutyrylglycerol, a triacylglycerol that was conveniently deprotected in ethanol with acrylic beads containing Candida antarctica lipase B to give [5 '',6 '',8 '',9 '',11 '',12 '',14 '',15 ''-H-3(N)]2-arachidonoylglycerol ([H-3]2-AG). The flash chromatographic separation necessary to isolate the labeled 2'-acylglycerol [H-3]2-AG resulted in only 4% of the rearrangement byproducts that have been a particular problem with previous methodologies. This reliable "kit" method to prepare the radiolabeled endocannabinoid as needed gave tritiated 2-arachidonoylglycerol [H-3](2)-AG with a specific activity of 200 Ci/mmol for enzyme assays, metabolic studies, and tissue imaging. It has been run on unlabeled materials on over 10 mg scales and should be generally applicable to other 2-acylglycerols.
