    Symmetric and Asymmetric Bolaamphiphiles from Ascorbic Acid

    The properties of novel bolaamphiphiles that carry epimers of vitamin C (L-ascorbic acid and/or D-isoascorbic acid) as hydrophilic head groups, and an interconnecting aliphatic C-12 chain (DD, DL, and LL) were investigated by differential scanning calorimetry (DSC), thermogravimetric analysis (TGA), small-angle X-ray scattering (SAXS), X-ray diffraction (XRD), and Fourier transform infrared spectroscopy (FTIR) in the solid state (anhydrous powders) and in aqueous dispersions as a function of the surfactant concentration. Upon heating, the aqueous dispersions undergo a phase transition from a hydrated semicrystalline "coagel" to a micellar phase. The results suggest that the headgroup chirality determines the formation of either inter- or intramolecular hydrogen bonds between the polar heads, which affect the phase behavior and structural properties of the nanoassemblies produced by these surfactants in water dispersions. The DSC data of aqueous dispersions were analyzed to obtain the size distribution of the pores in the coagel state.

    Polyhydroxylated hybrid molecules have been synthesized using a protocol based on the regioselective acylation of the target compounds with activated dicarboxylic acids catalyzed by Novozym-435. The procedure implies that the mixed ester derivatives prepared and isolated from the first esterification step act as acylating agents in the second esterification step.

  • Chemo-Enzymatic Synthesis of Silybin and 2,3-Dehydrosilybin Dimers

    Divalent or multivalent molecules often show enhanced biological activity relative to the simple monomeric units. Here we present enzymatically and chemically prepared dimers of the flavonolignans silybin and 2,3-dehydrosilybin. Their electrochemical behavior was studied by in situ and ex situ square wave voltammetry. The oxidation of monomers and dimers was similar, but adsorption onto the electrode and cell surfaces was different. A 1,1-diphenyl-2-picrylhydrazyl (DPPH) and an inhibition of microsomal lipoperoxidation assay were performed with same trend of results for silybin and 2,3-dehydrosilybin dimers. Silybin dimer showed better activity than the monomer, while on the contrary 2,3-dehydrosilybin dimer presented weaker antioxidant/antilipoperoxidant activity than its monomer. Cytotoxicity was evaluated on human umbilical vein endothelial cells, normal human adult keratinocytes, mouse fibroblasts (BALB/c 3T3) and human liver hepatocellular carcinoma cell line (HepG2). Silybin dimer was more cytotoxic than the parent compound and in the case of 2,3-dehydrosilybin its dimer showed weaker cytotoxicity than the monomer.

    本发明公开了曲克芦丁酰胺类衍生物及其制备方法和用途,属药物化学领域。该方法以曲克芦丁乙烯酯为原料,在离子液体中,反应温度40℃~70℃,分别与1,2,3,4‑四氢异喹啉类化合物或苯乙基胺、4‑氟‑苯乙基胺、苄胺化合物反应,得到曲克芦丁酰胺类衍生物,此类化合物具有抗肿瘤活性。本发明选离子液体既做溶剂又做催化剂合成曲克芦丁酰胺类衍生物,反应条件温和、操作简单,合成成本低,产率较高,为黄酮类化合物的结构修饰提供一条新的途径。
