Continuous Flow Synthesis of Secondary Amides by Tandem Azidation- Amidation of Anilines

The continuous flow synthesis of a variety of secondary amides by tandem azidation-amidation of anilines is described. This new procedure benefits from the improved safety feature of generating aromatic azides in flow, thus ensuring low concentrations of any potentially hazardous intermediates. The protocol was amenable to the production of multi-gram quantities of the amide product.

In this paper we report the scope and limitations of Pd(0)-mediated coupling reactions between aromatic iodides linked to a polystyrene resin and terminal acetylenes and olefins (Heck reactions). Optimized reaction conditions were evaluated with a number of different reagents. The optimized reaction conditions were frequently found to be superior to those previously reported in the literature and resulted in excellent yields of the products upon cleavage from the solid phase.

An iron(III)-promoted hydroalkynylation of unactivated alkenes toward Csp–Csp3 bond formation has been developed. Various alkenes, including mono-, di-, and trisubstituted alkenes, could all smoothly convert to structural diversified alkynes in this chemoselective protocol. Additionally, the scalability was unraveled and the further divergent transformations of products were conducted to demonstrate
