Abstract A convenient and efficient method for the synthesis of 1,5‐benzodiazepines by the reaction of o‐phenylenediamines (OPDA) and ketones with hydrogens at the α‐position has been developed. The reaction is carried out at room in the presence of a catalytic amount of magnesium perchlorate under solvent‐free conditions.

o-Phenylenediamines undergo smooth condensation with ketones having hydrogens at α-position on the surface of heteropoly acid (Ag3PW12O40) under extremely mild conditions to afford the corresponding 1,5-benzodiazepines in excellent yields with high selectivity. The catalyst can be recovered by simple filteration and can be reused in subsequent reactions.
Ceric ammonium nitrate was found to be an efficient reagent for the preparation of 1,5-benzodiazepine derivatives of a wide range of substituted o-phenylenediamines and electronically divergent ketones in moderate to excellent isolated yields (60-98%) under mild conditions using methanol as solvent at ambient temperature.
