In order to synthesize four stereoisomers of cystathionine (CYT), D- and L-homocysteines (D- and L-Hcy) were synthesized from methionine (Met) by a facile procedure. L-Met was reacted with dichloroacetic acid in concentrated hydrochloric acid under reflux to give (4S)-1,3-thiazane-2,4-dicarboxylic acid hydrochloride [(4S)-TDC.HCl]. L-Hcy was obtained by treatment of (4S)-TDC.HCl with hydroxylamine
为了合成胱硫醚(CYT)的四种立体异构体,通过简便的方法从蛋氨酸(Met)合成了D-和L-高半胱氨酸(D-和L-Hcy)。使L-Met与二氯乙酸在浓盐酸中在回流下反应,得到(4S)-1,3-噻嗪-2,4-二羧酸盐酸盐[(4S)-TDC.HCl]。通过用羟胺处理(4S)-TDC.HCl获得L-Hcy。还通过(4R)-TDC.HCl中间体由D-Met合成了D-Hcy。在碱性条件下,将获得的D-和L-Hcy与(R)-和(S)-2-氨基-3-氯丙酸盐酸盐缩合,得到四种CYT立体异构体。
