  摘要：

  The preparation of a series of 1-glutarylindoline-2(S)-carboxylic acid derivatives, 6a-v and 21a-c, is described. The above compounds were tested for inhibition of angiotensin converting enzyme. The structure-activity relationship of the series is also discussed. Compound 6u, the most potent member of the series, had an in vitro IC50 of 4.8 X 10(-9) M. Compound 6v, an ethyl ester of 6u, lowered blood pressure 70 mm in spontaneous hypertensive rats at an oral dose of 30 mg/kg.

  Die Erfindung betrifft hypotensive, antihypertensive und bradvkardische Verbindungen der Formel I worin Ph unsubstituiertes, 1,2-Phenylen oder durch einen bis drei, gleiche oder verschiedene Substituenten der Gruppe Niederalkyl, Niederalkoxy, Niederalkylendioxy, Hydroxy, Halogen oder Trifluormethyl substituiertes 1,2-Phenylen bedeutet, Ro für Wasserstoff oder HPh steht, jedes der Symbole R1, R2 und R3 Wasserstoff oder Niederalkyl bedeutet, und n für eine ganze Zahl von 1 bis 10 steht; ihre Amide, Mono- oder Di-niederalkylamide, Niederalkylester, (Amino, Mono- oder Di-niederalkylamino, Carboxy oder Carboniederalkoxy)-niederalkylester, und ihre Salze. Sie können z.B. durch Hydrolyse einer Verbindung der Formel worin mindestens eines der Symbole X and Y Cyan bedeutet, und das andere die gennante freie, amidierte oder veresterte Carboxygruppe ist, hergestellt werden.
