A structure-activity relationship investigation for a more efficacious therapy to treat onychomycosis, a fungal infection of the toe and fingernails, led to the discovery of a boron-containing small molecule, 5-fluoro-1,3-dihydro-1-hydroxy-2,1-benzoxaborole (AN2690), which is currently in clinical trials for onychomycosis topical treatment.

A Suzuki-type cross-coupling of aryldioxaborolane with 2-bromo-N,N-dimethylacetamide in the presence of a catalytic amount of tricyclohexylphosphine as the ligand and hydroquinone as the free-radical scavenger has been demonstrated as a convenient and simple way for the synthesis of alpha-arylacetamide. A simple, efficient, and environmentally benign boricacid catalyzed methodology for the ipso-hydroxylation of arylboronicacid to phenol has been developed using aqueous hydrogen peroxide as an oxidizing agent and ethanol as the solvent. The versatility of this protocol is that the reactions were performed at room temperature in short reaction time under metal-, ligand-, and base-free conditions.
