    8-Polycycloalkyl-1,3-dipropylxanthines as potent and selective antagonists for A1-adenosine receptors
    摘要:
    With the aim of characterizing the hydrophobic interactions between xanthines and the A1 receptor site, 1,3-dipropyl-8-substituted xanthines were synthesized. Introduction of a quaternary carbon and the conformationally restricted cyclopentyl moiety into the 8-position of xanthines enhanced the adenosine A1 antagonism. 1,3-Dipropyl-8-(3-noradamantyl)xanthine (42) was identified to be a selective and the most potent A1 receptor antagonist reported to date. Under our structure-activity relationship, the 8-substituent of xanthine antagonists and the N6-substituent of adenosine agonists appears to bind to the same region of the A1 receptor.

    Novel cyclopentane derivatives which are useful as an intermediate for the preparation of N-(L-aspartyl)-N'-(2,2,5,5-tetramethylcyclopentanecarbonyl)-R-1,1-diaminoe thane are represented by the formula (I) ##STR1## wherein R.sub.1 is hydrogen and R.sub.2 is cyano, formyl, carbamoyl, alkanesulfonyloxy, or ##STR2## wherein R.sub.3 and R.sub.4 are the same or different and represent alkyl, substituted alkyl or R.sub.1 and R.sub.2 are combined to be methylsulfinylmethylene, acetoxymethylthiomethylene, halomethylthiomethylene, methylthiomethylene, alkoxy-methylene, or --CH.sub.2 --O--.

    Process for the production of cycloalkanecarboxylic acids of general Formula I ##STR1## wherein n means the number 1 or 2, R.sub.1 and R.sub.3 are alkyl groups of 1-4 carbon atoms, and R.sub.2 and R.sub.4 symbolize hydrogen atoms or alkyl groups of 1-4 carbon atoms, characterized by reacting a compound of general Formula II X--CR.sub.1 R.sub.2 --(CH.sub.2).sub.n --CH.sub.2 --CR.sub.3 R.sub.4 --X'(II), wherein n, R.sub.1, R.sub.2, R.sub.3 and R.sub.4 have the meanings given above and X and X' mean a hydroxy group, a chlorine atom, a bromine atom, an alkylsulfonyloxy residue, or an arylsulfonyloxy residue, with 1,1-dichloroethylene; splitting off hydrogen chloride from the thus-obtained compounds of general Formula III X--CR.sub.1 R.sub.2 --(CH.sub.2).sub.n --CH.sub.2 --CR.sub.3 R.sub.4 --CH.sub.2 --CCl.sub.2 X (III), wherein n, X, X', R.sub.1, R.sub.2, R.sub.3 and R.sub.4 have the meanings given above; and cyclizing and hydrolyzing, by means of acids, the thus-prepared compounds of general Formula IV X--CR.sub.1 R.sub.2 --(CH.sub.2).sub.n --CH.sub.2 --CR.sub.3 R.sub.4 --CY.dbd.CZX' (IV), wherein n, X, X', R.sub.1, R.sub.2, R.sub.3 and R.sub.4 have the meanings given above, and Y and Z jointly represent a carbon-to-carbon bond or mean a hydrogen atom and a chlorine atom.

    Compounds of the formula: wherein n is 0 or 1, R is lower alkyl (substituted or unsubstituted), R' is H or lower alkyl, and R" is a branched alkyl, alkyl-cycloalkyl, cycloalkyl, polycycloalkyl (poly = 2 or more, fused or non-fused), phenyl or alkyl-substituted phenyl, and physiologically acceptable cationic and acid addition salts thereof, which compounds are potent sweeteners. These derivatives of gem-diaminoalkanes are many times sweeter than sugar and are free from undesirable flavor qualities. Furthermore, they possess an unanticipated high degree of solubility compared with known synthetic sweeteners. In addition, the compounds possess high stability so that they can be used in all types of beverages and in conventional food processing. Sweetening compositions and sweetened edible compositions of these compounds are also provided.
