    我们合成了七种新的KRN7000类似物:RCAI-133和154–159。RCAI-154、156和158是次级酰胺类似物,分别在直链C18酰基链的α位有一个羟基、一个甲基或两个甲基。RCAI-155、157和159是对应的N-甲基化的三级酰胺类似物,而RCAI-133是N-甲基化的KRN7000。其中,RCAI-133的PBS溶液在体内诱导小鼠淋巴细胞产生Th2偏向细胞因子,而RCAI-154至159显示出仅微弱或几乎无免疫刺激活性。因此,N-甲基化使糖脂主要产生Th2型细胞因子,而酰基α取代则对活性有害。

    Indole and indoline derivatives of the formula: wherein indicates a double bond or a single bond between carbon atoms, R1 represents an alkyl group containing 1 to 6 carbon atoms (which may be substituted by one or more than one of the same type of substituents selected from the hydroxy group, alkenyl groups containing 2 to 5 carbon atoms, and alkanoyloxy groups containing 2 to 7 carbon atoms) or a hydrogen atom, and one of the symbols R2 represents an alkyl group containing 6 to 24 carbon atoms and the other symbol R2 represents an alkyl group containing 1 to 24 carbon atoms, or a hydrogen atom, and R3 reptesents an alkyl group containing 1 to 6 carbon atoms or a hydrogen atom, are new compounds possessing useful pharmacological properties. They are of use in the treatment of diabetes mellitus, hyperlipoproteinaemic states, of atherosclerosis, and of associated disorders. Pharmaceutical compositions containing the compounds are described and so are numerous processes for oreparing the compounds.
