smoothly with trimethylphosphite in the presence of methyltriphenylphosphonium iodide to give N-acyl-2-(dimethoxyphosphoryl)glycinates in good or very good yields. The dimethoxyphosphorylglycinates may be isolated by column chromatography, or used directly for the Wadsworth–Emmons synthesis of α,β-dehydro-α-amino acids in a one-pot procedure without purification.

The syntheses of the naturallyoccurring unusual unsaturated aminoacid (S)-2-amino-3-methylene-4-pentenoic acid and the corresponding gamma-glutamyl dipeptide are described. Key steps are an N-acyliminium ion addition using allenyhnethylsilane as nucleophile and an enzymatic resolution mediated by the L-aminopeptidase from Pseudomonas putida.

The synthesis of the methyl esters of N-methoxycarbonyl- and N-benzyloxycarbonylaziridine-2-carboxylic acid 2 by reacting methyl 2-chloro-N-carbalkoxyglycinate 1 with diazomethane is described. The aziridines were readily converted to derivatives of O-methylserine 5 and S-methylcysteine 6.

Our research on hydantoin based TNF-α converting enzyme (TACE) inhibitors has led to an acetylene containing series that demonstrates sub-nanomolar potency (Ki) as well as excellent activity in human whole blood. These studies led to the discovery of highly potent TACE inhibitors with good DMPK profiles.
triphenylphosphonium tetrafluoroborate in the presence of ethyl azodicarboxylate and triphenylphosphine in THF at room temperature gives the respective alkyltriphenylphosphonium salts in good yields. The reaction also worked for the conversion of N-acyl-2-hydroxyglycinates into N-acyl-2-triphenylphosphonioglycinates.
