    Two general synthetic approaches to a novel series of 2H-[1]benzopyrano[3,4-b]pyridines are described together with their receptor binding profile at a variety of monoamine receptors in mammalian brain tissue. The biologically active members of this series fall into into one of two broad classes: 3,4,4a,5-tetrahydro-2H-[1]benzopyrano[3,4-b]pyridines or trans-1,3,4,4a,5,10b-hexahydro-2H-[1]benzopyrano[3,4-b]pyridines. By appropriate pharmacophoric modification potent selective ligands for D2, alpha-2, 5HT1A, and 5HT2 receptors may be obtained. The previously published in vivo data on certain key representatives of these series are also summarized.

    Die Erfindung betrifft Verbindungen der Formel I, worin X Sauerstoff oder Schwefel bedeutet, Ring A unsubstituiert oder durch einen bis drei gleiche oder verschieden Substituenten aus der Gruppe Hydroxy, Hydroxy-C1-7-alkyl, verethertes Hydroxy, verethertes Hydroxy-C1-7-alkyl, Acyloxy, Acyloxy- C1-7-alkyl, Halogen, C1-7-Alkyl, Trifluormethyl, Amino, Mono- oder Di-C1-7-Alkylamino und Acylamino substituiert ist, oder Ring A durch ein C1-7-Alkylendioxy substituiert ist. R Wasserstoff, C1-7-Alkyl oder Aryl-C1-7-alkyl bedeutet, R1 für Wasserstoff, C1-7-Alkyl, C1-7-Alkylthio-C1-7- alkyl, Amino, Acylamino, (Amino, Mono- oder Di-C1-7-alkyl-(emino)-C1-7-alkylcarbamoyl steht, sowie R2 bis R5 Wasserstoff oder C1-7-Alkyl bedeuten, ferner deren Dehydroderivate mit einer Doppelbindung in 1,2-Position oder in 1,10b-Position, wobei in letzterem Fall R5 nicht vorhanden ist, und deren Salze. Sie lassen sich als psychoaktive Mittel zur Behandlung von Krankheiten des ZNS verwenden.
