Activation by the enzyme or just a good leaving group? The guanidinophenyl (OGp) ester is a key activating group in substrate mimetics and chemoenzymatic peptidebondformation. We show that the leaving group ability of the ester is at least as important in this process as the specific interactions of the activating group with the enzyme.
