[reaction: see text] A 1:1 mixture of the platinum bis(phosphine) complex [(S)-4]PtCl2 [(S)-4 = (S)-3,5-t-Bu-4-MeO-MeOBIPHEP] catalyzes the intramolecular asymmetric hydroarylation of 2-(4-pentenyl)indoles in moderate to good yield with up to 90% ee.

are disclosed. Compositions comprising the compounds and methods for using the compounds to inhibit HCV are also disclosed.
Dibutyl zirconocene-mediated trans-selective cyclization of methoxyallyl-ene followed by the reaction with electrophiles is a convenient method for the synthesis of functionalized cyclopentane derivatives. Ring-closing metathesis of the obtained cyclopentane derivative yielded a common building block for the synthesis of sesquiterpene analogues with unnatural trans-[3.4.0]bicyclo-nonane skeleton.

Naphthalene-linked P2-P4 macrocycles within a tri-peptide-based acyl sulfonamide chemotype have been synthesized and found to inhibit HCV NS3 proteases representing genotypes 1a and 1b with single digit nanomolar potency. The pharmacokinetic profile of compounds in this series was optimized through structural modifications along the macrocycle tether as well as the P1 subsite. Ultimately a compound with oral bioavailability of 100% in rat, and a long half-life in plasma was obtained. However, compounds in this macrocyclic series exhibited cardiac effects in an isolated rabbit heart model and for this reason further optimization efforts were discontinued. (C) 2017 Elsevier Ltd.
