  The total synthesis of (-)-aspergilazine A, an alkaloid possessing a rare N1' to C6 bisindole bond, is described. A palladium-catalyzed N-arylation was used to selectively install the N1'-C6 bond in the presence of three other possible arylation sites. The ligand XPhos displayed a unique capability to efficiently carry out the N-arylation while simultaneously suppressing epimerization of the sensitive C9 stereocenters. This total synthesis has confirmed that aspergilazine A is a dimer of brevianamide F.

  (EN) The present invention is directed to conopeptides having 6-45 amino acids, including one or more bromo-tryptophan residues. More specifically, the present invention is directed to conopeptides having the general formula: R-(Cys)n-R1-B-R2-Cys-R3, wherein R is a peptide chain of 0-24 amino acids, R1 is a peptide chain of 0 to 31 amino acids, R2 is a peptide chain of 0-29 amino acids, R3 is a peptide chain of 0 to 26 amino acids, B is 6-bromo-tryptophan, n is 0 or 1 and the total length of the conopeptide is from about 6 to about 45 amino acids. The invention also includes pharmaceutically acceptable salts of the conopeptides. These bromo-tryptophan containing conopeptides invention are useful as antihelminthic agents, anti-vomiting agents, sleep-inducing agents, adjuncts to anesthesia, anticonvulsant or neuroprotective agents.(FR) La présente invention se rapporte à des conopeptides comportant de 6 à 45 acides aminés, et notamment au moins un reste de bromo-tryptophane. Plus spécifiquement, la présente invention se rapporte à des conopeptides représentés par la formule générale: R-(Cys)n-R1-B-R2-Cys-R3, où R est une chaîne peptidique comportant de 0 à 24 acides aminés, R1 est une chaîne peptidique comportant de 0 à 31 acides aminés, R2 est une chaîne peptidique comportant de 0 à 29 acides aminés, R3 est une chaîne peptidique comportant de 0 à 26 acides aminés, B est un 6-bromo-tryptophane, n est égal à 0 ou 1 et la longueur totale du conopeptide est comprise entre 6 et 45 acides aminés.
