  ( S )-Lifitegrast (LFT) 是新型整合素拮抗剂，经美国食品和药物管理局批准，用于治疗干眼病的体征和症状。文献中缺乏外消旋 LFT 的合成、制备和分析对映体分离以及手性互变研究。因此，在我们的研究中，LFT外消旋体的合成、对映体的手性制备纯化程序以及综合分析进展的重点是快速对映选择性分离和pH依赖性手性互变研究。 LFT外消旋体的合成采用2-氨基-3-(3-(甲磺酰基)苯基)丙酸盐酸盐和2-(苯并呋喃-6-羰基)-5,7-二氯-1,2,3,4-四氢异喹啉-以6-碳酰氯为起始原料。通过制备型手性 HPLC 从外消旋体中分离出 ( R )-LFT，并使用 Q-TOF、FT-IR、NMR 光谱和手性 HPLC 进行表征。 ( R )-LFT 的纯度测定为对映体过量为 99.12%。以水和甲醇为流动相，在 Chiralpak IC [固定在纤维素上的三(3,5-二氯苯基氨基甲酸酯)]
