用途

4-溴-2-硝基苯甲酸叔丁酯可用作医药合成中间体。该物质可通过4-溴-2-硝基苯甲酸与2-溴-2-甲基丙烷反应制备，进而用于制备化合物2-氨基-4-溴苯甲酸叔丁酯。后者同样广泛用作医药合成中间体。

制备

将5.0g 4-溴-2-硝基苯甲酸溶于50mL N,N-二甲基乙酰胺中，加入41g 碳酸钾、4.6g 氯化苄基三乙基铵和69mL 2-溴-2-甲基丙烷，并在55℃下搅拌10小时。冷却至室温后，再加入12mL 2-溴-2-甲基丙烷并继续在55℃下搅拌4小时。将反应混合物冷却至室温，然后加入水和乙酸乙酯进行萃取。分离有机层，并依次用10％柠檬酸水溶液和饱和氯化钠水溶液洗涤后，使用无水硫酸镁干燥并在减压下蒸发溶剂。向残留物中加入甲醇，通过过滤分离得到3.0g 4-溴-2-硝基苯甲酸叔丁酯，为白色固体。

1H-NMR (CDCl₃) δ：1.55（9H, s），7.63（1H, d, J=8.3Hz），7.77（1H, dd, J=8.3, 1.9Hz），7.95（1H, d, J=1.9Hz）。
