  Benzoylpyridines and benzophenones were synthesized and evaluated in vitro as p38alpha inhibitors and in vivo in several models of rheumatoid arthritis. Oral activity was found to depend upon substitution: 1,1-dimethylpropynylamine substituted benzophenone 10b (IC50: 14 nM) and pyridinoyl substituted benzimidazole 17b (IC50: 21 nM) showed highest efficacy and selectivity with ED50S of 9.5 and 8.6 mg/kg po in CIA.   A correlation was found between electronic properties and regioselectivity in the Friedel-Crafts reaction of substituted phenylacetic and benzoic acids with anisole, utilising substoichiometric (down to 1 mol%) amounts of catalyst. Relative reactivities of selected catalysts for this reaction were also studied.

  本发明提供一种二芳基酮的合成方法，包括如下步骤：采用苄基偕二乙酸酯与芳基锌试剂反应，原位得到乙酸二芳基甲酯；真空条件下去除溶剂，并向步骤S1的产物中加入四氯乙烷、二氯二氰苯醌和蒸馏水，在温度为80‑130℃条件下反应5‑12h；旋蒸除去溶剂，通过柱层析法分离得到二芳基酮产物。本发明提供的二芳基酮的合成方法，采用一锅两步法合成二芳基酮，反应原料简单易得，反应过程无需使用过渡金属催化剂，且反应条件温和，底物适用性好。
