  本发明属于天然药物领域，公开了一种肿瘤靶向性藤黄酸类化合物及其制备方法和用途。该化合物具有如式(I)所示的结构。其中，G为藤黄酸类化合物；P为低聚化乙二醇或其一端氨基化的衍生物；FA为叶酸或叶酸类似物；FA以酯键或酰胺键与P偶联，P以端羟基与G的羧基偶联。通过体外MTT检测实验，发现其对肿瘤细胞株(HeLa和HepG2细胞)具有更好的抑制效果，比藤黄酸类药物强4‑10倍，但对正常肝细胞LO2的毒性低6倍以上。该叶酸修饰的藤黄酸类化合物具有叶酸受体介导靶向肿瘤细胞的特性，可克服藤黄酸类抗癌药物选择性低和易多耐药性的问题，达到通过细胞凋亡、诱导自噬等途径清除癌细胞的效果。G‑P‑FA(I)。
