Amide α,β-Dehydrogenation Using Allyl-Palladium Catalysis and a Hindered Monodentate Anilide

A practical and direct method for the alpha,beta-dehydrogenation of amides is reported using allyl-palladium catalysis. Critical to the success of this process was the synthesis and application of a novel lithium N-cyclohexyl anilide (LiCyan). The reaction conditions tolerate a wide variety of substrates, including those with acidic heteroatom nucleophiles.
The disclosure provides processes for synthesizing Compound I, and pharmaceutically acceptable salts thereof.

bis(diimidazolylidene) NHC ligands and two non coordinating bromide counter ions in tetradentate C4 fashion. A survey of their catalytic activity in Buchwald−Hartwig amination has been performed. The newly synthesized complexes also catalyzed the amination of aryl chlorides in the presence of KOtBu. Various aryl chlorides and amines can react smoothly to give the corresponding aminated products in moderate to high yields
