    pH-responsive polymeric micelle based on PEG-poly(β-amino ester)/(amido amine) as intelligent vehicle for magnetic resonance imaging in detection of cerebral ischemic area

    A series of pH-responsive polymeric micelles is developed to act as intelligent carriers to deliver iron oxide (Fe(3)O(4)) nanoparticles and respond rapidly to an acidic stimuli environment for magnetic resonance imaging (MRI). The polymeric micelle can be self-assembled at physiological pH by a block copolymer, consisting of a hydrophilic methoxy poly(ethylene glycol) (PEG) and a pH-responsive poly(beta-amino ester)/(amido amine) block. Consequently, the Fe(3)O(4) nanoparticles can be well encapsulated into polymeric micelles due to the hydrophobic interaction, shielded by a PEG corona! shell. In an acidic environment, however, the pH-responsive component, which has ionizable tert-amino groups on its backbone, can become protonated to be soluble and release the hydrophobic Fe(3)O(4) nanoparticles. The Fe(3)O(4)-loaded polymeric micelle was measured by dynamic light scattering (DLS), superconducting quantum interference device (SQUID) and a 3.0 T MRI scanner. To assess the ability of this MRI probe as a pH-triggered agent, we utilize a disease rat model of cerebral ischemia that produces acidic tissue due to its pathologic condition. We found gradual accumulation of Fe(3)O(4) nanoparticles in the brain ischemic area, indicating that the pH-triggered MRI probe may be effective for targeting the acidic environment and diagnostic imaging of pathologic tissue.     The invention relates to highly biocompatible or biophilic un-cross-linked or cross-linked polymers comprising one or more side-chain active acrylic amino acids of formula I wherein: X is —NH(CH 2 ) 4 —, —O—C 6 H 4 —CH 2 —, —OCH 2 —, —O—CH(CH 3 )—, —S—CH 2 —, —O-proline, and R is H or CH 3 ; and wherein the polymer further includes a free radical initiator and, optionally, a cross-linking agent having a plurality of polymerizable ethylenically unsaturated groups. The invention further concerns various highly biocompatible, cross-linked co-polymers comprising one or more monomers of formula I, and one or more other polymerizable monomers. Uses of such polymers and co-polymers for the production of contact lenses, intraocular lenses, implants, wound healing slabs, additives for food and cosmetics, conductive plastics, spinnable fibers, and the like are disclosed.

    2-chlorosulfinyl-azetidin-4-ones of the formula wherein R is the residue of a carboxylic acid and R is a carboxylic acid protecting group, are prepared by reacting a penicillin sulfoxide ester of the formula wherein R and R1 are as defined above, with an N-chlorohalogenating agent, e.g. N-chlorophthalimide, in ar, inert organic solvent, e.g. toluene, under essentially anhydrous conditions in the presence of a cross-linked polyvinylpyridine polymer, e.g. poly-(4-vinylpyridine), said polymer containing between about 1 percent and about 10 percent cross-linking with e.g. divinylbenzene. The 2-chlorosulfinylazetidin-4-ones are used as intermediates for the preparation of 3-exomethylenecepham-4-carboxylic acid ester sulfoxides.
