Condensation of Vilsmeier Salts, Derived from Tetraalkylureas, with α-Hydroxy Amide Derivatives: One-pot Approach to Synthesize 2-Dialkylamino-2-oxazolin-4-ones
A novel and straightforward synthetic protocol was developed to synthesize 2-dialkylamino-2-oxazolin-4-ones from various Vilsmeier salts and α-hydroxy amides derivatives. Notably, thozalinone (3a), as a mild stimulant in tristimania and anorexic, could be synthesized simply and in a high yield using this methodology.

Another objective of the present invention is to provide novel methods for producing α-ketoamide reductases that reduce 2-chlorobenzoyl formamide to (R)-2-chloromandelamide with high optical purity, using NADPH as the coenzyme.

enzymes with 2-chlorobenzoyl formamide-reducing activity, and the biochemical properties of the purified enzyme were analyzed. The analysis of a partial internal amino acid sequence of the purified enzyme revealed that the enzyme may be encoded by the putative open reading frame (ORF) YDL124w reported in the genome analysis. YDL124w was cloned and expressed in
E. coli
, and was subsequently shown to encode the α-ketoamide reductase. It was found that these resulting transformants facilitated the production of (R)-2-chloromandelamide from 2-chlorobenzoyl formamide.
