Acetylenic ketones of type 4 and acetylenic acetals of type 6 (Scheme 1) are excellent cyclization precursors for the acid-catalyzed synthesis of various heterocycles. Using this methodology, 5- and 2-substituted 3-bromothiophenes of types 7 and 8 have been prepared in good to excellent yields.

A new and efficient method for the preparation of N-protected 5- and 2-substituted 3-bromopyrroles 15 and 16 via acid-catalyzed cyclization of the corresponding acetylenic ketones 6 and acetylenic acetals 14 has been found (Scheme 3). Using this methodology, rapid and quite general access to highly substituted pyrroles, which are known to exhibit a number of interesting biological activities, has been established.

Acetylenic acetals of type I(Scheme 1) and acetylenic ketones of type III(Scheme 1), 37 and 38(Scheme 7) are versatile synthetic precursors for the synthesis of various heterocycles by acid-catalyzed cyclization reactions.
