New synthesis of both D- and L-3-O-carbamoyl-2-deoxy-4-thioribosides, substrates for β-selective glycosylations

A new route to intermediates for the synthesis of 2′-deoxy-4′-thionucleosides is described. By utilising enantiomeric erythro-dibenzyldithioacetals, bothD- and L-3-O-(N-acyl)carbamoyl thiosugars, which give β-selectiveglycosylations, are radily produced.

THE identity of 2-deoxyribose as a carbohydrate constituent of thymus nucleic acid was demonstrated by Levene. Using acidic hydrolysis1 and enzymic methods2, it was possible to prove that 2-deoxyribose was combined with the purine residues in the nucleic acid, though the nature of the sugar combined with the pyrimidine moiety is still uncertain3.
