We have developed a facile but versatile method to introduce functional molecules into DNA on a CPG support. Threoninol, whose amino group was protected with an (allyloxy)carbonyl (Alloc) group, was introduced into DNA via the corresponding phosphoramidite monomer. After selective deprotection of the Alloc group by treatment with palladium(0), a dye with a carboxyl group could be introduced into the DNA on the CPG Support through an amide bond. The present teaching provides a fluorescent oligonucleotide probe having a high degree of design flexibility and wide applicability, as well as the use thereof.
