    Thermally stable GAD65 mutants are provided, which are useful for early diagnosis of insulin-dependent diabetes mellitus and retain antigenicity to pancreatic islet-β-cell-specific autoantibodies. Specifically, error-prone PCR is performed using a GAD65 gene as a template. A host microorganism is transformed with the thus obtained random gene library. Screening is performed using as an indicator the presence or the absence of antigenicity of the expressed protein to a pancreatic islet-β-cell-specific autoantibody after heat treatment. Thus, a GAD65 mutant having thermostability and antigenicity can be obtained.
