The present invention relates to a class of quinoline compounds that act as insulin secretagogues, i.e., induce insulin secretion from beta cells. The present compounds may be used to induce divalent calcium cation influx in pancreatic beta-cells or to activate voltage-dependent calcium channels in pancreatic beta-cells. The present invention further relates to compounds for use in the treatment of diabetes.
Bissulfonamide derivatives of formula (I) are capable of inhibiting: a) the biosynthesis of aromatic amino acids via the shikimate pathway and b) the catabolism of quinic acid, wherein: Ar is an aryl or heteroaryl group; R1 and R2 are the same or different and each represent hydrogen or alkyl or R1 and R2 together form a C1-C3 alkylene group, -CO- or -CS-; and R3 and R4 are the same or different and each represent -alkyl-aryl, -alkyl-heteroaryl, -alkenyl-aryl, -alkenyl-heteroaryl, -alkynyl-aryl-alkynyl-heterorayl, aryl or heteroaryl.

A versatile domino reaction that can be used for the synthesis of bicyclic benzo- or pyridyl-fused lactam and sultam derivatives is presented, enabling rapid synthesis of a variety of complex structures.

We report a C–Ncross-couplingreaction of weakly nucleophilic N-arylsulfonamides and arylbromides to access N,N-diarylsulfonamides using Ni(cod)(DQ) as a catalyst without additional ligands. The process is compatible with electron-deficient and electron-rich aryl and heteroaryl bromides (34 examples, 21–98%) and can be applied to the derivatization of an N-arylsulfonamide pharmaceutical compound
