  • Process for obtaining an organic acid from an organic acid ammonium salt, an organic acid amide, or an alkylamine organic acid complex

    Disclosed herein are methods for the recovery of an organic acid, such as a heat stable lactic acid, from a feed stream comprising at least one of an organic acid amide, an organic acid ammonium salt, or an alkylamine-organic acid complex. The feed stream that comprises the organic acid amide, organic acid ammonium salt, or alkylamine-organic acid complex is mixed with at least one azeotroping agent. The azeotroping agent is a hydrocarbon capable of forming at least one azeotrope with the organic acid that is produced by the thermal decomposition of the amide, ammonium salt, or complex in the feed stream. Preferably the azeotrope is a heteroazeotrope. The mixture comprising the feed stream and the azeotroping agent is heated to produce a vapor stream that comprises the azeotrope. The vapor stream can be condensed to a liquid stream, and the organic acid is recovered in the liquid stream that is produced. When the azeotrope comprising the organic acid is a heteroazeotrope, the vapor stream can be condensed into a liquid stream, which can be separated into a first phase and a second phase. The first phase contains the highest concentration of organic acid and the second phase comprises azeotroping agent. The organic acid can be further purified and/or concentrated from the separated first phase or from the liquid stream.

  • Production of medium chain length poly-3-hydroxy alkanoates in Escherichia coli, and monomers derived therefrom

    Production of medium chain length poly-3-hydroxyalkanoates (mcl-PHAs) in Escherichia coli is reported for the first time. To produce PHA in E. coli, a PHA polymerase encoding gene (phaC2) of Pseudomonas oleovorans GPo1 was introduced into various E. coli strains and fatty acid oxidation mutants thereof. Of these, E. coli fadA/B, E. coli fadRfadA or E. coli fadRfadB mutants, which are defective in specific steps of the fatty acid β-oxidation pathway, allowed production of mcl-PHAs from fatty acids ranging from octanoate to octadecanoate. Maximum amounts of about 10% PHA of total cell dry weight (w/w) were found. The predominant monomers found in these PHAs were always the same, independently of the chain length of the supplied fatty acids: 3-hydroxyoctanoate for even chain length fatty acids and a combination of 3-hydroxynonanoate and 3-hydroxyheptanoate for odd chain length fatty acids. Since the monomer composition of the produced polymer was independent of the fatty acid chain length, β-oxidation occurs in these β-oxidation mutants. The polymer may be hydrolyzed to yield interesting optically active monomers and their esters.
