Modified G:C pairs block the cleavage of DNA by restriction endonucleases, whereas small major‐groove modifications at A:T pairs are tolerated. A series of 7‐substituted 7‐deazaguanine 2′‐deoxyribonucleoside 5′‐O‐triphosphates (dGRTPs) have been synthesised, and their use as substrates for polymerase synthesis of modified DNA and their cleavage by REs have been studied.
