    Conjugates derived from non-steroidal anti-inflammatory drugs (NSAIDs) and methods of use thereof are disclosed, useful for, inter alia, identifying and localizing the site of pathology and/or inflammation responsible for the sensation of pain in a patient; for identifying the sites of primary, secondary, benign, or malignant tumors; and for diagnosing infection or confirming or ruling out suspected infection. The NSAID-based conjugates contain an imaging moiety. The conjugates concentrate at sites of increased cyclooxygenase expression, thus revealing the sites of increased prostaglandin production, which is correlated with pain and inflammation, and correlated with tumor presence and/or location. Identifying areas of increased COX expressing can also aid in screening for infections.

    Abstract The synthesis and labeling of 99mTc-N3-N′-[2-sulfanyl-ethylamino)acetyl]-2-aminoethyl-sulfanyl-1-hexanamide}thymidine (99mTc-NHT) were studied. In the presence of sodium glucoheptonate (GH) and ethylene diamine tetraacetic acid (EDTA), 99mTc-NHT was obtained by using bisaminoethanethiol (N2S2) as a bifunctional coupling agent.     本发明涉及医药领域,具体涉及一种靶向II型囊泡单胺转运体的分子探针及其制备方法和应用,本发明提供的靶向II型囊泡单胺转运体的分子探针,具有制备方法简便、快速的优势,且相对于核素11C、18F,99mTc作为SPECT显像核素可以由钼锝(99Mo‑99mTc)发生器淋洗而得,不必依赖医用回旋加速器,价廉易得。此外,本发明制备的靶向II型囊泡单胺转运体的分子探针具有良好的体外稳定性、亲脂性和生物学活性,对VMAT2具有较好的亲和力,具有对VMAT2进行示踪成像的潜力,可能进行脑部的VMAT2示踪成像。
  • 99mTc- and Re-labeled 6-dialkylamino-2-naphthylethylidene derivatives as imaging probes for β-amyloid plaques

    Based on the conjugate strategy, two neutral Tc-99m labeled 2-(1-(6-(dialkylamino)naphthalen-2-yl) ethylidene) malononitrile (DDNP) and 1-(6-(dialkylamino) naphthalen-2-yl) ethanone (ENE) derivatives, and their corresponding rhenium complexes were synthesized. In vitro fluorescent staining indicated that the corresponding rhenium derivatives selectively stained the beta-amyloid (A beta) plaques in the brain sections of AD model mice with low background. Compared with FDDNP and FENE, the affinities of the corresponding rhenium derivatives to A beta aggregates decreased about 10-14-fold. In vivo biodistribution experiments in normal mice showed that Tc-99m-MAMA-ENE displayed medium initial brain uptake (0.65 % ID/g at 2 min) with a reasonable washout from the brain (0.19 % ID/g at 2 h) while (99mTc)-MAMA-DDNP showed a low brain uptake (0.28 % ID/g at 2 min). Further optimize these Tc-99m-labeled tracers in order to improve their binding affinities to A beta plaques and diffusion through the blood brain barrier may generate useful imaging agents for SPECT.     Two uncharged Tc-99m-labeled phenylbenzoxazole derivatives were biologically evaluated as potential imaging probes for beta-amyloid plaques. The Tc-99m and corresponding rhenium complexes were synthesized by coupling monoamine-monoamide dithiol (MAMA) and bis(aminoethanethiol) (BAT) chelating ligand via a pentyloxy spacer to phenylbenzoxazole. The fluorescent rhenium complexes 6 and 9 selectively stainined the beta-amyloid plaques on the sections of transgenic mouse, and showed high affinity for A beta((1-42)) aggregates (K-i = 11.1 nM and 14.3 nM, respectively). Autoradiography in vitro indicated that [Tc-99m]6 clearly labeled beta-amyloid plaques on the sections of transgenic mouse. Biodistribution experiments in normal mice revealed that [Tc-99m]6 displayed moderate initial brain uptake (0.81% ID/g at 2 min), and quickly washed out from the brain (0.25% ID/g at 60 min). The preliminary results indicate that the properties of [Tc-99m]6 are promising, although additional refinements are needed to improve the ability to cross the blood-brain barrier.
