Spirocyclic Dihydropyridines by Electrophile-Induced Dearomatizing Cyclization of N-Alkenyl Pyridinecarboxamides
摘要:
On treatment with acylating or sulfonylating agents, N-alkenyl pyridine carboxamides (N-pyridinecarbonyl enamines) undergo a dearomatizing cyclization initiated by pyridine acylation and followed by intramolecular trapping of the resulting pyridinium cation. The products are spirocyclic dihydropyridines which may be further elaborated to spirocyclic heterocycles with drug-like features.

Both aldimines and ketimines are allylated under simple Barbier-type conditions using allylbromide and commercial magnesium foil or zinc dust in tetrahydrofuran at room temperature.
A zeolite-immobilised Lewis acid e.g. copper catalyst for performing carbonyl-ene and iminoene reactions is described. The catalyst can readily be separated from the reaction mixture and re-used in further reactions with minimal reduction in activity.

From the oxygen esters containing sulfur α-substituent, corresponding stannous enolates were prepared by the use of stannous trifluoromethanesulfonate. The enolates reacted with various imines to provide β-lactams.
