Provided herein are substituted fused oxazoline derivatives and substituted pyran derivatives useful as inhibitors of the HKII enzyme. The invention further provides pharmaceutical compositions of the compounds of the invention. The invention further provides medical uses of substituted fused oxazoline derivatives and substituted pyran derivatives, for example, as antitumor agents.
Allylation sulfur substituted active methylene compounds such as RSCH2Y [Y = C(=O)R, COOR, CN] with allylic bromides in the presence of bases gives a mixture of the SN2-product and the apparent SN2′-product. The former product is selectively obtained in alkylation in a nonpolar solvent. Synthetic utility of this regiospecific allylation is demonstrated in the simple synthesis of pellitorine.

The reaction of the allylic carbanion derived from 1-phenylthio-2-octene with conjugated cyclopentenone derivatives in the presence of hexamethylphosphoric triamide (HMPA) regioselectively gave the 1,4 adduct at the α carbon of the allylic sulfide. The reaction was employed for the synthesis of prostaglandin E1.
