    Might the observed α2A-adrenoreceptor agonism or antagonism of allyphenyline analogues be ascribed to different molecular conformations?

    We recently reported that the alpha(2)-adrenoreceptor (AR) ligand allyphenyline (9) significantly enhanced morphine analgesia (due to its alpha(2C)-AR agonism), was devoid of sedative side effects (due to its alpha(2A)-AR antagonism), prevented and reversed morphine tolerance and dependence. To highlight the molecular characteristics compatible with this behaviour and to obtain novel agents potentially useful in chronic pain and opioid addiction management, the allyl group of 9 was replaced by substituents of moderate steric bulk (MR) and positive or negative lipophilic (pi) and electronic (sigma) contributions in all the possible combinations. Effective novel alpha(2C)-agonists/alpha(2A)-antagonists (2, 3, 10, 12, and 17) were obtained. This study also demonstrated that contradictory combinations of the physicochemical parameters were similarly able to induce the alpha(2A)-activation. Since we had previously observed that the absolute configuration affected only the potency, but not the functional profile of the ligands, we hypothesized that the alpha(2A)-activation was governed by a ligand preferred conformation. From a structural overlay investigation it emerged that an extended conformation appeared to be associated with dual alpha(2C)-agonism/alpha(2A)-antagonism, whereas a folded conformation associated with alpha(2C)-/alpha(2A)-agonism.     A general selective synthesis of β-, γ- and δ-hydroxyethenyl ethers, a class of compounds containing two mutually reactive functionalities positioned at an interacting distance, is based on the reaction of diols with 1,2-bis-(phenylsulfonyl)ethylene (BPSE) followed by reductive elimination of the resulting β-phenylsulfonyl acetals with sodium amalgam.

    A latex having a reduced amount of 4-phenylcyclohexene is obtainable by a process comprising reacting A) 30-90% by weight of at least one ethylenically unsaturated monomer; B) 70-10% by weight of a diene; C) 1-10% by weight of α,β-unsaturated carboxylic acids, carboxylic acid nitriles, carboxylic acid amides, or mixtures thereof; and D) an auxiliary, an additive or mixtures thereof; wherein the sum of A, B and C is 100% by weight. The reaction follows a gradient regime for components A and B. In the gradient regime, an amount added per unit time of one of components A or B continuously increases, while simultaneously an amount added per unit time continuously decreases for one of components A or B which does not undergo the continuous increase; with the proviso that a starting molar ratio of A to B is adjusted from a range of 0.15 - 0.95 or 1.05 - 6.66 through at least one discontinuous change in the amount added per unit time to a target molar ratio of A to B, in the range of 1.05 - 6.66 or 0.15 - 0.95, and thereafter the change in the amount added per unit time is made i) constantly for A and B, and/or ii) decreasingly for A and ascendingly for B, and/or iii) decreasingly for B and ascendingly for A, in any sequence, individually or in combination.
