制备方法与用途

合成方法

7-氮杂吲哚加入到10 mL二氯甲烷中,在0℃下缓慢滴加4.2 mL冰乙酸,搅拌20分钟后,再逐滴滴入液溶解在20 mL二氯甲烷中的溶液。反应在0℃下进行2小时。通过薄层色谱法跟踪检测反应进程,直至未见反应物时停止反应。使用甲醇作为展开剂的TLC检测显示产物Rf值为0.57。随后,向反应体系中加入饱和亚硫酸钠溶液至淀粉KI试纸不再变蓝。浓缩二氯甲烷后,再加入20 mL,并在0℃下用NaOH调节pH至>13。将反应混合物溶剂旋干,然后用二氯甲烷溶解,用洗两次,有机相用无硫酸干燥。通过硅胶柱层析色谱分离,产物真空干燥后得到目标产物1H-吡咯[2,3-B]吡啶-6-

将1H-吡咯[2,3-B]吡啶-6-与液按一定比例加入到装有磁力搅拌器、温度计和回流冷凝管分器的四口圆底烧瓶中,再加入沸程为60~90℃的石油醚。反应在氮气保护下进行。通过薄层色谱法跟踪检测反应进程,直至未见反应物时停止反应。将混合液移至抽滤瓶,在40~70℃下进行减压蒸馏,除去低沸点的胺和溶剂后得到1H-吡咯[2,3-B]吡啶-6-胺
