    A convenient and rapid assay for activity and density of living cells in many samples is provided. That is, a oxidized type quinone such as 1,4-naphthoquinone, 2-methyl-1,4-naphthoquinone or tetramethyl-p-benzoquinone is reacted with a system to be determined. The Hydrogen peroxide thus produced is indirectly reacted with the colorimetric reagent by using enzyme or directly with the colorimetric reagent, the intensity of the color development thus caused is determined with a spectro phtometer(a colorimeter). A proper enzyme of the acceptor of hydrogen peroxide is peroxidase, indirect color reagent is sodium salt of N-(carboxymethylaminocarbonyl)-4,4'-bis(dimethylamino)-diphenylamine or leucocrystal violet are suitable for indirect colometric reagent, and direct colorimetric reagent is cytochrome c peroxidase.

    The present invention provides an assay method comprising the steps of: (a)contacting a phthalimide with a developing agent to produce a chemiluminescent molecule or a chemiluminescent substrate; and (b)producing the chemiluminescent molecule or chemiluminescent substrate. The assay method can be, for example, a method of detecting a target or detecting migration of a phthalimide from a first location to a second location. The phthalimide is preferably 3-aminophthalimide or 5-aminophthalimide and the phthalimide is suitably a component of a complex comprising the phthalimide and a molecule such as a peptide, oligopeptide, polypeptide or antibody. The molecule to which the phthalimide is linked can be a target binding agent having a binding affinity for a target or be a cellular probe. Detection of a target may also include determining the amount of target as an assay result. The amount of chemiluminescence may be indicative of the amount of target.

    A method of detecting a virus having a hemagglutinin-esterase activity in a sample is disclosed, the method includes: contacting the sample with a substrate for an enzyme of hemagglutinin-esterase (HE) of coronavirus or hemagglutinin-esterase fusion protein (HEF) of influenza C virus; and detecting activity of the enzyme, the detection of the activity indicates that the sample contains coronavirus or influenza C virus.
