    The hit-to-lead optimization of 1,2,3,4,4a,9a-hexahydro-1H-xanthenes as glucocorticoid receptor antagonists
    摘要:
    The structure activity relationship (SAR) study of a 1,2,3,4,4a,9a-hexahydro-1H-xanthene series of selective, human glucocorticoid receptor alpha (hGR alpha) antagonists is reported. Compounds were screened using hydroxyapatite-based GR binding and MMTV-Luc co-transfection reporter gene assays. Four different regions of the scaffold were modified to assess the effects on hGRa antagonism and related potency. Compound 8d exhibits an 8-fold better bioactivity than the original hit 1a, as well as an improved chemical stability, which make it a promising lead for the subsequent optimization.     Polyamide-polyurea containing polymers are disclosed that are derived from the reaction of (A) an organic polyisocyanate: (B) an organic compound having at least two active hydrogen containing groups; and (C) from 15 to 160 parts per 100 parts of said (B) of a chain extender combination comprising (i) a particular enamine and (ii) an aromatic diamine and optionally (iii) an extender diol wherein the ratio of equivalents of (A) to total equivalents of (B) and (C) is within the range of from 0.90:1 to 1.10:1. The polymers find particular utility in the preparation of RIM parts having excellent physical properties. At the same time, the polymers are characterized by slower gel times than prior art aromatic diamine extended polymers thereby allowing the filling of larger molded parts.

    Hydrocarbonaceous fuels and additive compositions therefor which comprise: a) one or more fuel-soluble manganese carbonyl compounds; b) one or more fuel-soluble alkali or alkaline earth metal-containing neutral or basic detergent salts; and c) one or more fuel-soluble ashless dispersants. These compositions preferably contain, in addition to components a), b) and c) above, one or more of the following: d) at least one fuel-soluble demulsifying agent; e) at least one aliphatic or cycloaliphatic amine; and f) at least one metal deactivator. The compositions possess improved combustion characteristics (e.g., formation of less soot, smoke, carbonaceous products and/or noxious emissions), and form on combustion carbonaceous products of reduced acidity. The deposition of sludge on critical engine or burner parts or surfaces is reduced and the fuels have improved stability and demulsibility characteristics. And the fuel compositions can result in decreased fuel consumption in diesel engines.
