  本发明公开了一种核苷磷脂类化合物及其化学合成方法和在核酸递送中的应用。所述核苷磷脂化合物为亲水的核苷酸头部及疏水的甘油烷基醚构成的阴离子型两亲性分子，结构如下通式I、3i或3j所示。该类分子中的核苷碱基可以与目标核酸的碱基形成氢键及发生π‑π堆叠作用；疏水尾链与核酸疏水性缀合结构间可发生范德华力作用。其自身可以在水溶液中组装形成超分子纳米结构，结合寡核苷酸后形成的纳米复合物可有效穿透细胞膜，实现负载的核酸胞内释放并发挥其生物学功能。本发明提出的核苷磷脂类化合物作为新型载体能高效转染核酸，在基因治疗领域具有极为广阔的应用前景。
